My own choice is TrueCrypt -- it's open source, secure, and available for a variety of platforms.

In my case, I have a 32Gb partition on my portable drive that's only accessible via TrueCrypt. Once it's running and available, it works like any other drive -- the encryption is transparent and (usually) almost instantaneous.

You DO need admin rights to run it, so it might not work for you on a work PC -- but the same will be true of most similar programs.

Note that the TrueCrypt download is the same whether you want to install it or use it portably. In the latter case, you can tell it just to extract itself somewhere, and it's then up to you to copy its various important components to the root of your flash drive so they're available wherever you take it.

Just how secure you want it to be is up to you -- but you can even go as far as to implement a deniable encrypted volume without too much difficulty, following the instructions in the included manual.
